Sperm Analysis & Sample Preparation

The analysis of the semen (sperm diagram) is performed with a microscopic and macroscopic examination.

The most important parameters defined in a sperm diagram are the following:

Volume ≥ 1.5 ml
pH ≥ 7.2
Liquefaction or not within an half hour Yes
Viscosity Normal
Shade of Colour The sample must not necessarily have a particular shade of colour.
Agglutination A normal sample should not exhibit agglutination. Nevertheless, if agglutination exists at a high percentage (over 50%) then it would be advisable to perform a control of the antibodies
Sperm concentration/ml ≥ 15 x 106
Total Sperm concentration/ml ≥ 39 x 106
Sperm mobility within an hour ≥ 40%
Round cells < 1 x 106
Morphology of the cells (according to Kruger) ≥ 4% normal forms (strict criteria)

Some of the terms we use to describe the state of the sample are the following:

  • Oligozoospermia (oligospermia) when the sperm concentration is less than 15 x 106

  • Asthenozoospermia (asthenospermia): when the sperm motility is less than 40%

  • Teratozoospermia (teratospermia): when the sperm morphology is below 4%

  • Azoospermia: when no spermatozoa can be detected in the sample.

Note: A sperm diagram serves to give us a first impression with regard to the sperm sample. In certain cases, it is necessary to repeat the examination since there is the possibility fluctuations in the sample parameters.
